Summary:

In this thesis, a new approach that provides high resolution 2D and 3D panoramic imaging with the control of the optimum range for cytopathological analysis processes in light microscopic systems is proposed. The proposed approach consists of four main stages: determining optimum range, creating 2D 3D adjacent images, controlling optimum range and 2D 3D panoramic imaging. In this study, Extended Depth of Field and Shape From Focus approaches are used for creating 2D 3D adjacent images. Literature studies that scan across the range with random start and end points on the Z axis to create 2D image sequence with the same field of view and different focuses for Extended Depth of Field and Shape From Focus approaches. In the first stage, the optimum range is estimated on the Z axis to obtain 2D and 3D images with maximum focus. In the second stage, new focus measurement operators are proposed for Extended Depth of Field and Shape From Focus approaches. In the third stage, the optimum range control is performed at every X or Y step of the platform, thus preventing the focus difference between 2D 3D adjacent images. In the final stage, two different panoramic imaging approaches are proposed, based on 2D images and hybrid. The proposed approach is compared with the studies in the literature and it is proved subjectively and objectively that it provides better results.
