Summary:

      In this study, 19 populations belonging to 9 wild Primula L. (Primulaceae) taxa were investigated with respect to nrDNA ITS regions.

Plant materials used in this study were collected from North Anatolia during the field work in 2005. Vouchers were identified firstly according to traditional methods and stored as herbarium specimens. Genomic DNA’s were isolated from healthy leaves of each populations to perform ITS studies. ITS regions of the examined populations were amplified by using universal primers and then sequenced. These ITS bands were aligned to use analysis processes. All ITS sequences were analyzed by using PAUP 4012b software and parsimony tree were formed in order to explain relationships among populations. Molecular evidences inferred from sequencing data were compared with morphological results in order to explore the exact relationships among the examined populations.

      This study suggest that to understand the phylogenetic relations among populations, ITS sequence analysis gives more reliable data compared to morphological characteristics.
