Summary:

      In this study, 19 populations belonging to D. orientale Hoffm. (Asteraceae) were compared based on nrDNA-ITS and RAPD profiles. Plant materials used in this study were collected in suitable vegetation period during the field study in 2004-2010. DNA isolation was carried out from healthy silica-dried leaves. Primarily, ITS regions of the examined populations were amplified by using ITS universal primers and then sequenced. It was determined that length of ITS region varies between 703-711bp. While length ITS1 varies between 209-211 bp, length of ITS2 varies between 257-258 bp. In addition, it was found that % GC contents of whole ITS varies between 53,2-53,6. While, % GC content is 53,1-53,6 in ITS1 it is 51,8-52,7 in ITS 2. The size of DNA bands obtained using three RAPD primers varies from 350 to 2000 bp. It was found that 21 of total 23 bands were polimorphic. Examined samples base on the bands profile were grouped according to similarity level. Additionally molecular variations among the examined populations were identified by phylogenetic analysis.
