Summary:

      The aims of this study are to determine the best germination medium for T. pseudopulegioides seeds, as well as to produce seedlings via direct organogenesis from the explants being placed on the Murashige & Skoog (MS) basal media, each supplemented with various plant growth regulators and determine total phenolics and and antioxidant activities of the extracts obtained from aforesaid plantlets. Explants (ca. 1 cm long) were cut from the seedlings which were previously grown on MS with PGR free medium and they were immediately placed on MS basal media supplemented with 6-BA, TDZ and KIN with the concentrations of 0,25; 0,5; 1,0 and 2,0 mg/L, individually. After two months culturing, methanol extracts of seedlings were evaluated for their total phenolic and flavonoid contents as well as their antioxidant activities. The former was carried out by using HPLC, while the latter was evaluated by two complementary test systems, namely DPPH free radical scavenging and β-Carotene color bleaching test. Total phenolic contents of all samples ranged from 24,01 to 1083,74 (mg/100 g) and whilst those of flavonoids were from 22,48 to 92,08 (mg/100 g). The highest total phenolic content was observed in medium including 1,0 mg/L KIN at the range of 1083,74 mg and the highest total flavonoid content was observed in medium including 0,5 mg/L 6-BA at the range of 92,08 mg, respectively. The highest free radical scavenging activity was observed of 2,0 mg/L KIN. In β-Carotene color bleaching test the highest activity observed of 0,5 mg/L 6-BA.
