Summary:

In this study, 51 populations belong to 34 Paronychia Mill. (Caryophyllaceae) taxa, including endemic 20 taxa for Turkey, were investigated in terms of nrDNA ITS data. Healthy leaf samples used in DNA isolation was collected during the field work of a project supported by TUBITAK (111T820) in 2012-2013. ITS regions of the examined populations were amplified using universal primers and sequencing by Macrogen Inc. It was determined that length of ITS region varies from 635 to 671 bp, GC % content is between 59.2-62.3, the ratio of Purine / Pyrimidine varies from 1.027 to 1088. The aligned sequence data of nrDNA ITS region belong to examined taxa contain 142 parsimonik (20 %), 230 variable (32 %) and 431 conservative (invariant) site (59 %). It was also found that P.argentea var. argentea and P. kayseriana are the farthest taxa with 75.7 % similarity level, P. davisii, P. adalia and P. lycica are the closely related taxa with 100% similarity level. Additionaly, according to GC % content, P. argentea var. scariarissima and P. carica var. stipulata are the farthest taxa, P. davisii, P. adalia, P. lycica, P. carica are the closely related taxa. As a result of phylogenetic analysis based on the nrDNA ITS data, Paronychia appears to be a polyphyletic genera.
