Summary: Chimeric xylanase genes were constructed from B. halodurans C-125 and Geobacillus sp. TF16 genes with a method based on DNA shuffling. These chimeric genes were named as GeoInH2CTer, GeoInH2CTer2, GeoIntraH2Int3 and GeoIntraH2Int4. Chimeric genes were cloned into pET28a(+) expression vector. GeoIntraH2Int3 and GeoIntraH2Int4 were succesfully expressed in E. coli BL21 (DE3), but there was no expression for GeoInH2CTer and GeoInH2CTer2. Optimum temperature of GeoIntraH2Int3 and GeoIntraH2Int4 were 60 °C, 50-60 °C and optimum pH of them were 8,0 and 8,0-10,0, respectively. Key words: Bacillus halodurans C-125, Geobacillus sp.
