Summary:

      In this study, to develop an effective microbial control agent against Acanthoscelides obtectus, we isolated bacteria and characterized as morphological, physiological, biochemical and molecular. In the studies to determine the bacteria from A. obtectus, 9 bacterial isolates have been isolated and named as Ao1-9. They are determined as morphological using light microscopy, biochemical features using both conventional microbiological methods and API kits. For molecular characterization, analyzed the sequences of 16S rDNA gene and compared the isolates in GenBank. According to the obtained results, 7 of these isolates are Gram (+) (Ao1, 2, 3, 4, 5, 8 ve 9), 5 of them include spore form, (Ao3, Ao4, Ao5, Ao8 and Ao9), and 2 of isolates are Gram (–) (Ao6 ve Ao7). Oxidase enzyme activity was determined in 3 isolates (Ao6, Ao7 ve Ao8) as positive, other isolates were observed as oxidase negative. According to catalase enzyme activity test, 6 of isolates (Ao1, 2, 4, 5, 8 and 9) were observed as positive. And the rest of 3 isolates (Ao3, Ao6 and Ao7) were also observed as catalase negative. According to the results of 16S rDNA base sequence analysis, Ao5 isolate was identified as Bacillus thuringiensis. According to the insecticidal activity tests results, the most effective isolate was 80% for Ao5 isolate. B. thuringiensis isolate (Ao5) may be valuable as biological control agent against A. obtectus.
